my peugeot speed fighter 2 will not start?

The motorcycle will not start after some fault diagnostics . I have the following : Good spark ( new plug fitted ) Good fuel ( cleaned carb and confirmed fuel type and flow )Compression appears good .I am unable to get the engine to fire , even with engine start spray.

Have popped a small amount of fuel into chamber . have also tried start spray . not even a hint of a fire , after a few kicks the bike backfires .I have also tried to place hand over air intake with no luck .

take plug out and pop a little fuel down bore hole. replace plug and try to start does it fire?
