when you take coloured tab out of top of tap, whats supposed to happen when you undo the screw

After removing the screw you can then pull the handle piece away - usually on splines.

Search on Google Images for "faucet diagram" lots of exploded pictures of different types of tap.

If you continue after removing tap handle, make sure water is turned off - or call an expert.
