How can i stop a service valve from leaking?

Service valve to a WC (cistern) leaking at the WC end. The nut in the middle works but I can't seal it. Tried PTFE tape. Also can't turn off the surply water to repalace the hole valve.

I have turned the water off under the sink and also at the two gate valves in the loft by the Cold water cistern. There still seems to be pressure in the pipe which would stop me from taking the service valve off to replace it. I think the first time I tried I may have over tightened this. I have an Header tank in the loft so I think my system should be indirect. Very confused!

can you turn the water off , with a (mains key) from out side your home , there should be a mains off valve inside your home , near the sink, if not use the one outside your home on the foot path , , you can get a mains key in a hardware store ,
