I see on my bathroom (upstairs sink tap) there is a scew on the pipe below ?

is this an isolation point? also the tap won't work it simply is not coming on (cold tap). How do i fix it?

yes an isolation valve.
if its old be careful when you turn it they are prone to leak from the seal behind the screw
tap may have a faulty washer or jumper valve.
